Three basic types of fluorescence microscopy and recent improvement

چکیده

Fluorescence microscopy is one of the most used imaging modalities in molecular biology and living specimens. To increase image contrast spatial resolution, different type fluorescence has been developed. This review introduces three main types microscopy: wild-field microscopy, confocal total internal reflection microscopy. The basic principles are similar but with modifications, which also indicates their attributes limitation. recent improvement on these microscopies discussed. Some techniques show advance overcoming common microscopy's weakness, future perspectives

Three-dimensional holographic fluorescence microscopy.

Most commonly used methods for three-dimensional (3D) fluorescence microscopy make use of sectioning techniques that require that the object be physically scanned in a series of two-dimensional (2D) sections along the z axis. The main drawback in these approaches is the need for these sequential 2D scans. An alternative approach to fluorescence imaging in three dimensions has been developed tha...

Two-Photon Fluorescence Microscopy: Basic Principles, Advantages and Risks

The application of two-photon excitation to fluorescence microscopy has become a powerful tool for studying biological function in live tissue and offers many advantages over conventional imaging techniques. Neuroscientists in particular have used this technology to image physiological functioning in microscopic and subcellular neural compartments. Neurons can be imaged deep within highly light...

Three-photon excitation in fluorescence microscopy.

We show experiments proving the feasibility of scanning fluorescence microscopy by three-photon excitation. Three-photon excitation fluorescence axial images are shown of polystyrene beads stained with the fluorophore 2,5-bis(4-biphenyl)oxazole (BBO). Three-photon excitation is performed at an excitation wavelength of 900 nm and with pulses of 130 fs duration provided by a mode-locked titanium-...

Evaluating performance in three-dimensional fluorescence microscopy

In biological fluorescence microscopy, image contrast is often degraded by a high background arising from out of focus regions of the specimen. This background can be greatly reduced or eliminated by several modes of thick specimen microscopy, including techniques such as 3-D deconvolution and confocal. There has been a great deal of interest and some confusion about which of these methods is '...

Scattered light fluorescence microscopy in three dimensions.

Recently, we have proposed a method to image fluorescent structures behind turbid layers at diffraction limited resolution using wave-front shaping and the memory effect. However, this was limited to a raster scanning of the wave-front shaped focus to a two dimensional plane. In applications, it can however be of great importance to be able to scan a three dimensional volume. Here we show that ...
